RIPA裂解液 (RIPA Lysis Buffer) C1053
描述: RIPA裂解液(RIPA Lysis Buffer)對動物細胞胞膜、胞漿、胞核成分均有較強裂解作用,是經典和最常用的細胞組織快速裂解液。使用RIPA 裂解緩沖液制備用于Western特別是用于免疫共沉淀的裂解產物已經是一種首選的標準操作,也適用于大部分抗原表位檢測,特別是免疫共沉淀等實驗。
組成: 100 ml RIPA裂解緩沖液。
成分:50 mM Tris-HCl (pH 7.4),

注意:如所得蛋白產物較為粘稠,可先
制備組織裂解產物:
1. 取50-100 mg組織在冰上剪成碎片,用預冷的PBS洗滌2次離心棄去PBS;
2. 加入 0.5-1 ml 預冷 RIPA 裂解緩沖液;

注意:如所得蛋白產物較為粘稠,可先
說明:
1. 轉移上清液時不要吸入底部的沉淀物;
2. 在做免疫沉淀或免疫共沉淀時最好在實驗前進行蛋白提取,避免某些不穩定蛋白的降解;
3. RIPA裂解緩沖液中未加入蛋白酶抑制劑,用戶可自行選擇添加。
